Abstract Propene (propylene, PE) is an important industrial chemical which is also present as a contaminant in urban air and cigarette smoke. PE is metabolized to propylene oxide (PO), which binds to macromolecules, i.e. haemoglobin and DNA. PE is an asphyxiant. Human data are scarce. Inhalation of PE on a long-term basis gives rise to non-neoplastic toxic changes in the nasal cavity of rats, but not of mice. In male mice there is a higher incidence of chronic focal renal inflammation. In female mice there is a higher incidence of uterine endometrial stromal polyps and, to a lesser extent, haemangiosarcoma as well as haemangiosarcoma and haemangioma combined. There are limited data on the potential health hazard to humans due to PE exposure. However, as a metabolite, PO is carcinogenic in experimental animals. It is not possible at the present time to rule out PE as a human carcinogen.
